That depends on the size of the quilt and the pattern that you use. Below are some examples if you just want to sew the squares together using a quarter inch seam allowance. You can use fewer squares by adding wide solid borders.

Queen Size (90" x 96") = 440 squares
Full/Double (84" x 90") = 380 squares
Twin/Single (69" x 90") = 320 squares
Crib/Lap Size (42" x 50") = 120 squares
